717解毒合剂对蝮蛇咬伤大鼠心肌NO、MDA、SH水平的影响Effect of 717 Antidote Mixture on the Levels of NO,MDA and SH in Myocardium of Rats Bitten by Agkis-trodon

摘要(Abstract):
目的 观察717解毒合剂对蝮蛇咬伤大鼠心肌一氧化氮(NO)、丙二醛(MDA)、巯基(SH)水平的改善作用。方法 8周龄SPF级SD大鼠50只,雌雄各半,随机分为正常对照组、模型组、717解毒合剂低剂量组(0.6mL/100 g)、717解毒合剂中剂量组(1.2 m L/100 g)、717解毒合剂高剂量组(2.4 mL/100 g)。除正常对照组外,其余大鼠均予2.5μg/g腹蛇毒液于左后肢皮下注射造模,并于造模成功2 h后开始给药,连续3 d。末次给药12 h后取大鼠心脏制备10%心肌组织匀浆,离心后提取上清并检测心肌MDA水平、心肌NO水平、心肌总巯基(TSH)水平、心肌非蛋白结合巯基(NPSH)水平和心肌蛋白结合巯基(PSH)水平。结果 与正常对照组相比,模型组大鼠心肌MDA水平和NO水平表现出显著升高的状态(P <0.05),心肌TSH、NPSH、PSH水平则呈现降低的状态(P <0.05);中药治疗组大鼠心肌MDA水平和NO水平显著下调(P <0.05),心肌TSH、NPSH、PSH水平明显上调(P <0.05),并呈现剂量依赖性趋势,给药剂量越高,心肌损伤的改善程度越优。结论 717解毒合剂对蝮蛇毒诱导的大鼠心肌损害具有一定的改善和保护作用,可以改善心肌MDA、NO、TSH、NPSH、PSH水平,值得进一步研究。
