Box-Behnken响应面法优化虎杖苷的提取工艺及体外活性评价Optimization of Extraction Process of Polydatin by Box-Behnken Response Surface Methodology and Invitro Activity Evaluation

摘要(Abstract):
目的优化虎杖中虎杖苷回流提取工艺,并评价其体外抗氧化与抑菌活性。方法采用单因素法考察温度、乙醇体积分数、液料比、提取时间对虎杖提取物中虎杖苷含有量的影响,以Box-Behnken响应面法优化提取工艺;采用DPPH法评价其抗氧化活性;采用牛津杯法测定最小抑菌质量浓度,并绘制抑菌生长曲线。结果虎杖苷的最佳提取工艺:温度77.54℃,乙醇体积分数60%,液料比16.53 m L/g,提取时间1.54 h;虎杖提取物对DPPH自由基具有良好的清除率,且对金黄色葡萄球菌、枯草芽孢杆菌均有抑制作用,并呈剂量依赖性。结论基于响应面法优化虎杖苷所得的提取条件稳定可靠,且虎杖提取物具有一定抗氧化和抑菌作用,对后续的深入研究具有指导意义。
