电刺激对脊髓损伤大鼠神经再生及NRG-1/ErbB-PI3K/Akt通路的影响Effects of Electrical Stimulation on Nerve Regeneration and NRG-1/Erb B-PI3K/Akt Pathway in Rats with Spinal Cord Injury

摘要(Abstract):
目的观察电刺激对脊髓损伤大鼠神经再生及NRG-1/ErbB-PI3K/Akt通路的影响。方法 SD大鼠60只随机分为3组:对照组、模型组、电刺激组。模型组、电刺激组建立脊髓损伤模型,建模成功后,电刺激组给予电疗刺激(强度为40μA,脊髓上的电场强度为400μV/mm,振荡周期持续时间15 min,每日1次,连续7周);对照组、模型组无任何处理措施。实验结束后测定各组大鼠机械痛阈(PWT)、热刺激潜伏期(PWTL)、脊髓损伤的行为学评分(BBB)、运动诱发电位(MEP)水平;随后处死大鼠,测定脊髓组织NRG1、ErbB、PI3K、Akt mRNA、蛋白表达水平。结果与对照组比较,模型组PWT、PWTL、BBB评分、NRG1、ErbB、PI3K、Akt mRNA、蛋白表达水平降低,MEP潜伏期升高(P <0.05);与模型组比较,电刺激组PWT、PWTL、BBB评分、NRG1、ErbB、PI3K、Akt mRNA、蛋白表达水平升高,MEP潜伏期降低(P <0.05);与对照组比较,电刺激组PWT、PWTL、BBB评分、NRG1、ErbB、PI3K、Akt mRNA、蛋白表达水平略微降低,MEP潜伏期略微升高,差异无统计意义(P> 0.05)。结论电刺激能减轻大鼠脊髓损伤程度,促进损伤神经再生;其机制与电刺激激活NRG-1/ErbB-PI3K/Akt通路,促进NRG1、ErbB、PI3K、Akt mRNA、蛋白的表达有关。
