地黄饮子汤对rt-PA溶栓后急性脑梗死大鼠神经细胞凋亡及Bcl-2、Caspase-3表达的影响Effect of Dihuang Yinzi Decoction on Neuronal Cell Apoptosis and Expression of Bcl-2 and Caspase-3 in Rats with Acute Cerebral Infarction after rt-PA Thrombolysis

摘要(Abstract):
目的观察地黄饮子汤对重组组织型纤溶酶原激活剂(rt-PA)溶栓后急性脑梗死(ACI)大鼠神经细胞凋亡及B细胞淋巴瘤-2(Bcl-2)、Caspase-3表达的影响。方法将40只大鼠用随机数字表法均分为4组:假手术组、模型组、重组组织纤溶酶原激活剂(rt-PA)组和地黄饮子汤组。采用注射鼠凝血酶法制备大鼠大脑中动脉栓塞模型,20 min后rt-PA组和地黄饮子汤组大鼠尾静脉注射rt-PA(10 mg/kg),地黄饮子汤组同时灌胃给予地黄饮子汤(5 g/kg),对照组和模型组给予等量生理盐水。24 h后处死大鼠,用TTC染色确定脑梗死体积,检测血清中白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)水平,用TUNEL法检测神经细胞凋亡情况,RT-PCR法检测脑组织中Bcl-2、凋亡基因(Bax)和Caspase-3 m RNA表达。结果与假手术组比较,其余各组大鼠脑梗死体积、细胞凋亡率、IL-6、TNF-α、Bax和Caspase-3水平增高,Bcl-2表达降低;与模型组比较,rt-PA组和地黄饮子汤组大鼠脑梗死体积、细胞凋亡率、IL-6、TNF-α、Bax和Caspase-3水平降低,Bcl-2表达增高;与rt-PA组比较,地黄饮子汤组大鼠脑梗死体积、细胞凋亡率、IL-6、TNF-α、Bax和Caspase-3水平降低,Bcl-2表达增高。结论地黄饮子汤可能通过增加脑梗死区Bcl-2表达,抑制Caspase-3表达,从而降低rt-PA溶栓后急性脑梗死大鼠神经细胞凋亡,为地黄饮子汤防治急性脑梗死溶栓后再灌注损伤提供了理论依据。
