金刚藤颗粒对急性盆腔炎模型大鼠NLRP3炎症小体通路及免疫功能的影响Effects of Jingangteng Granule oEffects of Jingangteng Granule on NLRP3 Inflammatory Body Pathway and Immunologic Function of in Rats with Acute Pelvic Inflammation

摘要(Abstract):
目的观察金刚藤颗粒对急性盆腔炎(APID)模型大鼠NLRP3炎症小体通路及免疫功能的影响。方法将雌性SD大鼠105只随机分为假手术组、模型组及金刚藤颗粒低、中、高剂量组,每组均15只,灌胃治疗15 d后,ELISA检测大鼠血清白细胞介素-1β(IL-1β)、白细胞介素-18(IL-18)水平;苏木精-伊红(HE)染色观察子宫组织的病理形态;实时定量PCR及Western blotting分别检测NLRP3、ASC、IL-1β、Caspase-1 mRNA及蛋白表达水平。结果 HE染色显示,模型组大鼠子宫内膜上皮细胞脱落坏死,腔壁结构紊乱,全层炎性细胞浸润,有充血水肿;与模型组比较,金刚藤颗粒治疗组症状较模型组有显著改善;与对照组、假手术组比较,模型组血清IL-1β、IL-18水平显著升高(P <0.05),与模型组比较,金刚藤颗粒治疗组IL-1β、IL-18水平显著降低(P <0.05);与对照组比较,模型组NLRP3、ASC、IL-1β、Caspase-1 mRNA水平显著升高(P <0.05),与模型组比较,金刚藤颗粒治疗组NLRP3、ASC、IL-1β、Caspase-1 mRNA水平显著降低(P <0.05);模型组NLRP3、ASC、IL-1β、Caspase-1蛋白表达水平显著高于假手术组(P <0.05),金刚藤颗粒治疗组NLRP3、ASC、IL-1β、Cas-pase-1蛋白表达水平较模型组显著降低(P <0.05)。结论金刚藤颗粒对急性盆腔炎模型大鼠具有显著治疗作用,其可能通过抑制NLRP3炎症小体通路发挥免疫作用。
