三七总皂苷在乙酸所致小鼠急性炎性内脏痛中的镇痛作用Analgesic Effect of Panaxnotoginsengsaponins on Acute Inflammatory Visceral Pain in Mice Induced by Acetic Acid

摘要(Abstract):
目的分析三七总皂苷(PNS)在乙酸所致小鼠急性炎性内脏痛中的镇痛作用。方法选取成年雄性昆明小鼠120只,随机分为对照组(n=20只),生理盐水组(n=50只),三七总皂苷组(n=50只)。对照组常规条件下饲养,不做干预建立急性内脏痛模型;生理盐水组预先腹腔注射生理盐水,剂量为2.86 m L/kg;三七总皂苷组预先腹腔注射三七总皂苷7 mg/mL,同时两组间隔12 h注射1次,持续7 d,第8日腹腔注射1%乙酸(10 mg/kg),建立急性内脏痛模型,即刻中观察昆明小鼠扭体反应。结果乙酸腹腔注射后3 min左右产生行为学反应,生理盐水组在15~30 min后扭体反应达到峰值,三七总皂苷组在30~45 min出现反应高峰值;三七总皂苷组和生理盐水组比较,0~15、15~30、45~60、60~75、120~135 min后扭体反应明显减弱(P <0.05);生理盐水组、三七总皂苷组与对照组比较显著增加(P <0.05);三七总皂苷组GFAP表达显著低于生理盐水组(P <0.05)。结论乙酸所致小鼠急性炎性内脏痛模型预先经腹腔注射三七总皂苷可抑制脊髓胶质细胞的激活,进一步缓解急性炎性内脏痛。
