身痛逐瘀汤对大鼠中重度退变腰椎间盘髓核内TNF-α、IL-1β的影响Effect of Shentong Zhuyu Decoction on TNF-α and IL-1β in Nucleus Pulposus of Lumbar Intervertebral Disc in Rats with Moderate to Severe Degeneration

摘要(Abstract):
目的观察身痛逐瘀汤对大鼠中重度退变腰椎间盘髓核内肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)的影响并探讨其机制。方法将128只雄性SD大鼠按照随机原则分为假手术组、模型组、西药组和中药组,每组32只。采用纤维环穿刺法建立腰椎间盘退变模型,造模成功后西药组、中药组、假手术组和模型组分别给予尼美舒利分散片、身痛逐瘀汤、等量0.9%氯化钠注射液灌胃,连续喂养20 d后检测灌胃后不同时间椎间盘髓核内TNF-α、IL-1β的表达。结果西药组和中药组在灌胃后椎间盘髓核内TNF-α、IL-1β表达与灌胃前比较差异具有统计学意义(P <0.05),两组比较差异无统计学意义(P> 0.05),此后两组炎性因子的表达呈缓慢上升趋势,西药组灌胃后2周与灌胃前比较差异无统计学意义(P> 0.05),而中药组与灌胃前比较差异有统计学意义(P <0.05),西药组和中药组灌胃后4周与灌胃前比较均差异均有统计学意义(P <0.05),中药组和西药组在灌胃后2周和4周髓核内TNF-α、IL-1β表达比较差异均有统计学意义(P <0.05)。结论身痛逐瘀汤能明显降低中重度退变椎间盘内TNF-α和IL-1β的表达,抗炎止痛效果稳定而持久。
