二冬膏对LPS诱导小鼠肺泡巨噬细胞分泌TNF-α、IL-6及AQP1的影响

摘要(Abstract):
目的观察二冬膏对脂多糖(LPS)诱导小鼠肺泡巨噬细胞分泌肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)及水通道蛋白1(AQP1)的影响,并探讨可能的作用机制。方法以二冬膏低、中、高剂量灌胃大鼠,采集二冬膏含药血清。以空白血清为对照,考察二冬膏含药血清对LPS诱导小鼠肺泡巨噬细胞MH-S的影响。MTT法检测MH-S细胞存活率,一氧化氮(NO)测定试剂盒检测NO的含量,ELISA和RT-PCR法检测TNF-α、IL-6及AQP1的水平,Western blotting法检测MH-S细胞中Toll样受体4(TLR4)、髓样分化因子(MyD88)、核转录因子κB p65(NF-κB p65)及AQP1的水平。结果二冬膏各剂量组在干预24 h内对MH-S细胞的存活率没有明显差异。与模型组相比,二冬膏含药血清能剂量依赖性地降低NO、TNF-α及IL-6的分泌量,明显抑制TNF-α和IL-6 mRNA的水平(P <0.05或P <0.01),显著促进AQP1 mRNA的表达(P <0.05或P <0.01);能明显下调TLR4、MyD88及NF-κB p65蛋白的水平(P <0.05或P <0.01),上调AQP1蛋白的水平(P <0.01)。结论二冬膏含药血清能够通过上调AQP1,抑制TLR4/MyD88/NF-κB信号通路,从而抑制炎性因子的表达及释放,发挥抗炎作用。
