三七总皂苷结合针灸干预对急性缺血性脑卒中大鼠血脑屏障的保护作用及对PI3K/Akt信号通路的影响Protective Effect of Panax Notoginseng Saponins Combined with Acupuncture on Blood-brain Barrier and the Effect on PI3/KAkt Signaling Pathway in Acute Ischemic Stroke Rats

摘要(Abstract):
目的观察三七总皂苷(PNS)结合针灸治疗急性缺血性脑卒中大鼠血脑屏障保护作用及对PI3K/Akt信号通路的影响。方法将大鼠随机分为假手术组、模型组、针灸组、PNS+针灸组4组,每组各15只。采用Zea Longa线栓法制作大鼠缺血性脑卒中模型,假手术组大鼠仅分离血管,而不插入栓线,针灸组及PNS+针灸组大鼠术后给予针灸治疗,PNS+针灸组大鼠在针灸治疗基础上尾静脉注射PNS,连续治疗7 d。比较大鼠神经行为学评分、血脑屏障通透性、脑组织含水量以及脑组织PI3K、Akt蛋白表达。结果术后24 h各脑卒中组大鼠神经行为学评分无显著差异(P> 0.05),术后3 d、术后7 d针灸组、PNS+针灸组大鼠神经行为学评分较模型组显著降低(P <0.05),且PNS+针灸组大鼠神经行为学评分显著低于针灸组(P <0.05)。各组大鼠左脑组织伊文思蓝(EB)含量、含水量无显著差异(P> 0.05),各脑卒中组大鼠右脑组织EB含量、含水量显著高于假手术组(P <0.05),其中针灸组与PNS+针灸组大鼠右脑组织EB含量、含水量较模型组显著降低(P <0.05),而PNS+针灸组大鼠右脑组织EB含量、含水量显著低于针灸组(P <0.05)。各脑卒中组大鼠脑组织PI3K、AKt表达水平均较假手术组显著升高(P <0.05),其中针灸组以及PNS+针灸组大鼠脑组织PI3K较模型组显著升高(P <0.05),且PNS+针灸组大鼠脑组织PI3K表达显著高于针灸组(P <0.05);而模型组、针灸组以及PNS+针灸组大鼠脑组织Akt表达差异无统计学意义(P> 0.05)。结论 PNS联合针灸治疗急性缺血性脑卒中大鼠具有显著的血脑屏障保护作用,可有效改善大鼠神经行为学状况,其作用机制可能与促进PI3K/Akt信号通路的激活有关。
