泻白散治疗急性肺损伤作用机制的网络药理学研究Exploration of Xiebai Powder on Mechanism in Treating Acute Lung Injury Based on Network Pharmacology

摘要(Abstract):
目的 应用网络药理学探讨泻白散治疗急性肺损伤(ALI)的分子机制。方法 通过文献检索及TCMSP在线数据库筛选泻白散的有效成分及靶点预测,从Genecards数据库获取ALI相关疾病靶点,用Venny图将药物靶点与疾病靶点进行交集,获取共同作用靶点。把共同作用靶点用Cytoscape 3.7.2软件进行蛋白互作网络构建及关键靶点筛选,通过Metascape数据库进行GO生物功能富集分析和KEGG通路,借助微生信进行可视化。结果 泻白散治疗ALI疾病的活性成分为“quercetin槲皮素”“kaempferol山柰酚”“naringenin柚皮素”等,核心靶点有NPM1、TP53、COPS5、MCM2、CUL1、CUL3;其主要生物学通路有MAPK信号通路、FoxO信号通路、cAMP信号通路以及JAK-STAT信号通路等,与调控炎症反应、抗氧化反应、介导细胞凋亡等功能密切相关。结论 本研究初步展示了泻白散治疗ALI疾病多成分、多靶点、多通路的作用机制,为泻白散的临床应用提供理论依据。
