熄风静宁颗粒对Tourette综合征模型大鼠脑纹状体多巴胺D2受体和多巴胺转运蛋白mRNA表达的影响Effects of Xifeng Jingning Granule on Dopamine Receptor D2 and Dopamine Transporter mRNA Expression in Striatum of Tourette Syndrome Rat

摘要(Abstract):
目的观察熄风静宁颗粒对Tourette综合征(TS)模型大鼠脑纹状体多巴胺D2受体(DRD2)和多巴胺转运蛋白(DAT)mRNA表达的影响。方法 Wistar雄性大鼠36只随机分为空白对照组、模型组、氟哌啶醇组、熄风静宁颗粒低剂量组、中剂量组、高剂量组,采用亚氨基二丙腈(IDPN)腹腔注射建立TS大鼠模型,造模后连续给药6周,采用RT–PCR法检测大鼠脑纹状体中DRD2和DAT mRNA表达的情况。结果 IDPN诱导的模型大鼠的体质量显著低于正常大鼠(P<0.01);给药前后熄风静宁颗粒中剂量组TS模型大鼠体质量增长与正常大鼠体质量的增长无差异;TS模型大鼠脑纹状体DRD2 mRNA与正常组比较呈现高表达(P<0.01);熄风静宁颗粒中、高剂量组和氟哌啶醇可降低TS模型大鼠脑纹状体DRD2 mRNA的表达(P<0.05);TS模型大鼠脑纹状体DAT mRNA表达与正常组比较显著增高(P<0.01);中药颗粒高剂量组能够降低TS模型大鼠脑纹状体DAT mRNA的表达,但氟哌啶醇作用更明显(P<0.05)。结论熄风静宁颗粒尚具有部分调节机体状态的作用;Tourette综合征存在多巴胺功能的亢进,熄风静宁颗粒能够通过降DRD2受体的敏感性,抑制多巴系统系统功能的亢进而发挥药物治疗的作用。
