人参皂苷Rg_5对胃癌BGC-823细胞增殖、凋亡及凋亡相关因子的影响The Effect of Ginsenoside Rg_5 on Proliferation,Apoptosis and Related Factors of Apoptosis of BGC-823 Cellin Gastric Cancer

摘要(Abstract):
目的观察人参皂苷Rg_5对胃癌BGC-823细胞增殖、凋亡及凋亡相关因子Bcl-2相关X蛋白(BAX)、B细胞淋巴瘤-2(Bcl-2)表达的影响,探讨人参皂苷Rg_5抑制胃癌生长转移的可能作用机制。方法采用CCK-8法检测人参皂苷Rg_5对BGC-823细胞24、48 h的细胞增殖抑制作用,流式细胞术检测高、低浓度人参皂苷Rg_5对BGC-823细胞凋亡的影响,Western blot法检测凋亡相关因子Bax、Bcl-2蛋白的表达。结果与空白组比较,人参皂苷Rg_550、100、200μmol/L 24 h存活率降低(P<0.05),人参皂苷Rg_512.5、25、50、100、200μmol/L 48 h存活率降低(P<0.05);与空白组比较,低浓度人参皂苷Rg_5凋亡率升高(P>0.05),高浓度人参皂苷Rg_5凋亡率升高(P<0.05);与空白组比较,高低浓度人参皂苷Rg_5组Bax蛋白表达均升高(P<0.05),高浓度人参皂苷Rg_5组Bcl-2蛋白表达降低(P<0.05)。结论人参皂苷Rg_5可有效抑制胃癌BGC-823细胞的增殖,促进BGC-823细胞凋亡,且其促进BGC-823细胞凋亡的机制可能与其降低线粒体途径相关的细胞凋亡因子Bcl-2蛋白的表达,升高Bax蛋白的表达相关。
