川芎嗪注射液通过抗炎、抗氧化和抗凋亡减轻脂多糖诱发急性肺损伤的研究Study of Ligustrazine Hydrochloride Injection on Alleviating Lipopolysaccharide-induced Acute Lung Injury by anti-inflammatory,antioxidant and apoptosis

摘要(Abstract):
目的观察川芎嗪注射液对急性肺损伤(ALI)的治疗作用并探讨其可能的机制。方法利用脂多糖(LPS)建立ALI体内体外疾病模型。测定促炎细胞因子的蛋白水平和一氧化氮(NO)含量,炎症介质和凋亡基因的表达水平,活性氧(ROS)含量,丙二醛(MDA)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-PX)的变化,观察肺脏病理改变及肺组织湿/干重(W/D)比值变化,及肺组织的细胞凋亡情况。结果川芎嗪注射液抑制LPS诱导的ROS生成和肿瘤坏死因子-α(TNF-α)、白细胞介素-β(IL-β)、白细胞介素-6(IL-6)的分泌,下调核因子E2相关因子(Nrf2)、血红素加氧酶-1(HO-1)、诱导型一氧化氮合酶(iNOS)、环氧化酶2(COX2)、髓样细胞触发受体-1 (TREM-1)、髓样细胞触发受体-2 (TREM-2)、核转录因子-κB p65 (NF-κB p65)、细胞间黏附分子-1(ICAM-1)和血管细胞黏附分子-1(VCAM-1)的表达,显著降低ALI细胞和大鼠模型中NO和MDA的水平,抑制SOD活性,增强GSH-PX活性,抑制肺细胞凋亡,抑制Bax增强Bcl-2的表达,并且川芎嗪注射液可明显减轻LPS所致的肺组织病理改变,降低肺组织W/D比值。结论川芎嗪注射液具有抗炎、抗氧化和凋亡的作用,能够减轻LPS导致的肺组织细胞损伤,起到较好的保护作用。
