肾病Ⅰ号方对单侧输尿管梗阻大鼠Wnt通路Wnt4和β-catenin表达的影响Effect of Nephropathy 1st Decoction on Expression of Wnt4 and β-catenin in Wnt Signaling Pathway of Unilateral Ureteral Obstruction Rats

摘要(Abstract):
目的研究肾病Ⅰ号方对Wnt通路中Wnt4和β-catenin表达的影响及肾病Ⅰ号方抗肾纤维化的机理。方法雄性SPF级SD大鼠108只,随机分为假手术组、模型组、西药组(盐酸贝那普利)、肾病Ⅰ号方高、中、低剂量组,每组18只。假手术组仅游离左侧输尿管,其余5组行左侧输尿管结扎术,于术后第2天灌胃。各组分别于灌胃后第7、14、21天随机选取6只大鼠留取24 h尿液,检测24 h尿蛋白(24 h UP);心脏取血,检测血肌酐(Scr)和尿素氮(BUN);处死大鼠,取肾脏组织行Masson染色,光学显微镜下观察肾脏组织病理改变;免疫组化方法观察Wnt4和β-catenin在肾间质的阳性表达情况。结果模型组和给药组大鼠血Scr和BUN较假手术组显著升高(P<0.05),24 h UP无统计学差异(P>0.05),Masson染色示蓝色胶原组织增多,Wnt4和β-catenin表达增加(P<0.05)。西药组和肾病Ⅰ号方高、中、低剂量组大鼠血Scr和BUN较模型组显著降低(P<0.05),24 h UP差异无统计学意义(P>0.05),Masson染色示蓝色胶原组织减少,Wnt4和β-catenin表达降低(P<0.05)。与西药组比较,肾病Ⅰ号方高、中、低剂量组大鼠各检测指标无统计学差异(P>0.05)。结论肾病Ⅰ号方可保护肾小管,并可能通过降低肾小管上皮细胞中Wnt4和β-catenin的表达,减少细胞外基质堆积,从而达到抗肾纤维化的作用。
