蒽醌类中药大黄与麦冬中大黄素的含量测定与比较研究The Experimental Studies on Determination and Comparison of Rhubarb and Radix Ophiopogonis in Anthraquinone Hherbs

摘要(Abstract):
目的建立蒽醌类中药大黄与麦冬中大黄素含量的测定方法以及两者间含量的比较。方法随机化分组方法将豚鼠分为大黄高、低剂量组及麦冬高、低量组,分别灌胃给予高、低不同剂量的麦冬及大黄后,采用高效液相色谱法测定大黄各剂量组和麦冬各剂量组豚鼠血浆中大黄素的含量,色谱柱为Stamsil-Bp C18(250 mm×4.6 mm,5μm),流动相为甲醇∶乙腈(60∶40,v/v),检测波长为434 nm。结果大黄高低、剂量组均可检测到大黄素(含量分别为5.69 mg/mL和3.58 mg/mL),麦冬高剂量组的含量为3.61 mg/mL,麦冬低剂量组中未检测到大黄素。大黄素血浆浓度在4~28μg/mL范围与峰面积线性关系良好,相关系数r=0.9996,平均回收率为92.05%~98.55%;日内及日间相对标准偏差分别为1.76%,2.43%。结论该方法简单、准确、可靠,灵敏,可有效测定大黄及麦冬中大黄素的含量。
