电针水沟对脑出血大鼠大脑皮质NPY调节作用的动态观察Dynamic Observation of the NPY Regulation Effect of Electroacupuncture Shuigou (DU26) in Rats with Cerebral Hemorrhage

摘要(Abstract):
目的观察神经肽Y(NPY)在脑出血大鼠大脑皮层神经细胞和内皮细胞中的动态表达,探讨电针水沟穴对脑出血大鼠脑血管神经调节物质NPY的作用及可能机制。方法将健康青年wistar大鼠随机分为正常组、假手术组、模型3、24、72h组、电针3、24、72h组,每组各10只。采用胶原酶Ⅶ诱发的大鼠尾壳核出血模型;电针组取水沟穴治疗,用连续波,频率120次/min,强度1mA,留针30min。造模麻醉醒后立即针刺1次,每隔24h再针刺1次。根据Bederson神经体征评分法进行综合等级评分,并采用免疫组化方法检测各组大鼠皮层组织NPY的动态表达。结果 (1)造模麻醉醒后电针治疗组的神经缺损体征综合评分略高于模型组。3d后,电针治疗组神经缺损体征综合评分与模型组及电针治疗前比较均有明显差异。(2)正常组与假手术组大鼠大脑皮层神经元和内皮细胞有少量的NPY阳性表达;脑出血后3h大鼠大脑皮层NPY的阳性表达开始增高,24h呈最高峰,胞浆和突起浓染呈棕褐色,血管内皮细胞细胞质中有棕色颗粒,72h时有所下降,与正常组比较均有显著差异。电针组大鼠大脑皮层神经元和内皮细胞的NPY阳性表达,也于3h开始表达,于24h表达较高,72h表达减弱,与模型组比较均有显著差异。结论电针水沟穴能有效抑制脑出血后3、24、72h时大脑皮质神经元和血管内皮细胞NPY的表达增强,改善脑出血大鼠的行为体征。
