膝痛方对木瓜蛋白酶诱导的兔膝骨关节炎模型IL-1β、TNF-α、MMP-3的影响Effect of Xitong Decoction on the Expression of IL-1β,TNF-α and MMP-3 in Rabbit Model of Knee Osteoarthritis Induced by Papain

摘要(Abstract):
目的观察膝痛方对骨关节炎急发期动物模型白介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、基质金属蛋白酶-3(MMP-3)的调控作用。方法 28只新西兰白兔随机分为空白组、模型组、扶他林组、膝痛方组,各7只,于第1、4、7日向右膝关节内注射2%的木瓜蛋白酶和0.03 mol/L L-半胱氨酸混合液0.5 m L(空白组除外)。末次注射7 d后开始治疗。治疗14 d后,测右膝局部皮温,取右膝关节髌下脂肪垫组织处滑膜组织0.3 g,HE染色进行组织病理学观察,ELISA测定滑膜匀浆IL-1β、TNF-α、MMP-3浓度。结果模型组右膝局部皮温略高于其他各组,但各组间差异并无统计学意义(P>0.05);与空白组比较,模型组滑膜增生明显,炎性细胞大量浸润,IL-1β、TNF-α、MMP-3明显升高(P<0.05);与模型组比较,膝痛方组及扶他林组滑膜增生较少,少量炎性细胞浸润,IL-1β、TNF-α、MMP-3降低(P<0.05);与扶他林组比较,膝痛方组IL-1β、TNF-α水平与之差异无统计学意义(P>0.05),而MMP-3则低于扶他林组(P<0.05)。结论膝痛方及扶他林均具有抑制膝骨关节炎急性期滑膜炎症反应的作用,且这种作用可能是通过降低IL-1β、TNF-α、MMP-3而实现的。此外,膝痛方抑制MMP-3的作用优于扶他林,提示膝痛方可能在远期抑制软骨退变,延缓膝骨关节炎病理进程方面具有潜在的疗效,仍需进一步研究。
