基于JAK/STAT3信号转导通路探讨黄芩苷预处理对糖尿病大鼠心肌缺血再灌注损伤的保护作用Effects of Baicalin Pretreatment on Diabetic Myocardial Ischemia-Reperfusion Injury in Rats Based on JAK/STAT3 Signal Transduction Pathway

摘要(Abstract):
目的观察黄芩苷对糖尿病大鼠心肌缺血再灌注损伤的保护作用。方法将雄性SD大鼠给予高脂饲料后腹腔给予链脲佐菌素的方法制备糖尿病模型,造模成功后将60只大鼠均分为假手术组、缺血再灌注组、AG490组、黄芩苷组和黄芩苷+AG490组。采用结扎左冠状动脉前降支的方法建立心肌缺血再灌注损伤模型。AG490组和黄芩苷组在结扎前20 min分别静脉给予AG490 1μmol/kg、黄芩苷100 mg/kg,黄芩苷+AG490组给予AG490 1μmol/kg+黄芩苷100 mg/kg,假手术组和缺血再灌注组给予等量生理盐水。恢复血流2 h后心脏取血,处死大鼠,检测心肌梗死面积、血清中LDH和CK含量以及心肌组织中JAK1、JAK2和STAT3 mRNA的水平。结果给予黄芩苷后,大鼠心肌梗死面积、血清中LDH和CK含量均降低,心肌组织中JAK1、JAK2和STAT3 mRNA相对表达水平增高;在给予了AG490干预后,大鼠心肌梗死面积、血清中LDH和CK含量均增加,心肌组织中JAK1、JAK2和STAT3 mRNA表达水平降低。结论黄芩苷能减小糖尿病大鼠心肌缺血再灌注损伤造成的心肌梗死面积,这可能与激活JAK/STAT-3信号转导通路相关。
