三七皂苷R1对血管平滑肌细胞迁移及p38MAPK蛋白的影响Effects of Notoginsenoside R1 on Vascular Smooth Muscle Cell Migration and p38 MAPK Protein

摘要(Abstract):
目的 观察三七皂苷R1对血管平滑肌细胞(VSMC)迁移及p38 MAPK蛋白的影响。方法 用AngⅡ诱导VSMC迁移建立细胞迁移模型,将迁移的VSMC随机分为对照组和三七皂苷R1低、中、高剂量组(5、10、20μmol/L)。用细胞划痕实验检测VSMC迁移面积,Western blotting法检测MMP2、MMP9、磷酸化p38 MAPK及p38 MAPK蛋白表达。结果 与对照组相比,三七皂苷R1中、高剂量组均能明显减少VSMC的细胞迁移面积及MMP2、MMP9迁移蛋白表达(P <0.05),且呈剂量依赖性;与对照组相比,三七皂苷R1中、高剂量组均能明显减少p38 MAPK蛋白磷酸化(P <0.05),且呈剂量依赖性。结论 三七皂苷R1抑制了AngⅡ诱导的VSMC迁移面积,同时下调MMP2和MMP9迁移蛋白,且呈剂量依赖性,其机制可能与降低p38 MAPK蛋白磷酸化有关。
