参麦注射液对急性束缚性应激大鼠大脑皮质Bcl-2/Bax表达及炎症因子的影响Influence of Shenmai Injection on Acute Immobilization Stress and BCL-2/BAX Expression and Inflammatory Factors in Rat with Cerebral Cortex

摘要(Abstract):
目的观察参麦注射液预处理对急性束缚性应激大鼠大脑皮质Bcl-2/Bax及炎症因子表达的影响,并探讨其作用机制。方法将24只雄性SD大鼠按随机数字表法分为正常对照组、参麦对照组、模型组、参麦预处理组,每组6只。采用专业大鼠固定器束缚大鼠2h来模拟急性应激状态。参麦预处理组于制模前30min、参麦对照组于相应时间腹腔注射参麦注射液15mg/kg;正常对照组和模型组则给予等量0.9%氯化钠注射液。采用酶联免疫吸附试验(ELISA)检测制模前、后血浆促肾上腺皮质激素(ACTH)和皮质酮(CORT)水平;制模后,采用实时荧光定量反转录-聚合酶链反应(RT-PCR)检测大脑皮质Bcl-2、Bax mRNA表达,蛋白质免疫印迹试验(Western Bolt)检测大脑皮质组织Bcl-2、Bax蛋白表达,采用酶联免疫吸附试验(ELISA)检测大脑皮质组织肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)表达水平。结果大鼠急性束缚性应激后血浆ACTH和CORT水平均明显高于应激前(P<0.01)。与正常对照组比较,模型组大脑皮质组织Bcl-2 mRNA表达明显降低,Bax mRNA和蛋白表达明显增高(均P<0.01),炎症因子TNF-α和IL-6表达量明显增加(均P<0.01)。与模型组比较,参麦预处理组大脑皮质组织Bcl-2 mRNA表达明显增高(P<0.01),Bax mRNA和蛋白表达明显降低(均P<0.01),炎症因子TNF-α和IL-6表达量明显降低(均P<0.01)。各组间大脑皮质Bcl-2蛋白表达差异均无统计学意义(P>0.05)。参麦对照组各指标影响与正常对照组比较差异均无统计学意义(P>0.05)。结论参麦注射液预处理可以维持急性束缚性应激大鼠皮质组织Bcl-2/Bax和炎症因子TNF-α和IL-6表达,从而缓解急性束缚性应激后的急性脑损伤。
