愈心梗液对急性心肌梗死大鼠心肌能量代谢和内皮功能障碍的影响The Influence of Yuxingeng Liquid (愈心梗液 ) on Myocardial Energy Metabolism and Endothelium Dysfunction in Rats with Acute Myocardial Infarctiion

摘要(Abstract):
目的观察实验性心肌梗死(AMI)大鼠心肌组织内皮素(ET)、血管紧张素II(AngII)水平的变化,并通过电镜观察心肌细胞线粒体(Mit)的形态结构并测定其ATP酶、琥珀酸脱氢酶(SDH)、酸性磷酸酶(ACP)、碱性磷酸酶(ALP)的活性和糖原(Gly)的含量,探讨愈心梗液对大鼠AMI后心肌血管活性物质和能量代谢的影响。方法结扎大鼠造成AMI模型,将大鼠随机分为空白组、穿刺组、模型组、开搏组及愈心梗液大、中、小剂量组,每组10只;各给药组于手术即日起开始灌胃给药,4周后处死大鼠,测定大鼠心肌组织及血中ET、AngII含量,电镜观察Mit的形态结构,组化染色及图象处理分析ATP酶、SDH、ACP、ALP的活性和Gly含量的改变。结果与模型组比较,大鼠服用愈心梗液后心肌组织及血浆中ET、AngII含量明显降低,开搏通组及愈心梗液大、中、小剂量组ATP酶、SDH、ACP、ALP的活性和Gly的含量显著升高。结论愈心梗液可显著改善AMI大鼠心肌组织内皮功能,并可提高心肌细胞酶活性和Gly含量,保护Mit结构的相对完整性,这可能是其防治AMI后早期心室重构(VR)的作用机理之一。
