灯盏花素对脑梗死大鼠脑保护作用的影像学观察及机制研究Imaging Observations and Mechanism Research of the Protective Effect of Breviscapine on Rats with Cerebral Infarction

摘要(Abstract):
目的采用核磁共振(MRI)技术,观察灯盏花素对脑梗死大鼠的保护作用并探索其机制。方法 120只SD大鼠随机分为空白组、模型组、尼莫地平组和灯盏花素组。除空白组外,其余各组动物脑制造脑梗死模型,尼莫地平组和灯盏花素组于梗死后每日腹腔注射尼莫地平1 mg/kg和灯盏花素4 mg/kg,模型组注射同等体积0.9%氯化钠注射液,连续4 d。缺血后24 h、48 h和96 h,用MRI对大鼠两侧纹状体区和顶颞叶区进行T1-SE及DW-EPI序列扫描,分析表观弥散系数(ADC)值的变化;并用干湿重法计算脑含水量及western法检测APQ4水平的变化。结果与模型组相比,灯盏花素组组大鼠脑缺血后24 h、48 h及96 h,左侧纹状体和顶颞叶相对ADC值著增加(P<0.01),脑含水量及APQ4值显著降低(P<0.01),而尼莫地平组和灯盏花素组相当(P>0.05)。结论灯盏花素对脑缺血再灌注损伤大鼠有一定的减轻脑水肿的作用,其机制可能与抑制APQ4的表达有关。
