参芎化瘀胶囊通过MAPK/ERK对脑缺血/再灌注大鼠的神经保护作用研究Neuroprotective Effect Study of Shenxiong Huayu Capsule Through MAPK/ERK on Cerebral Ischemia/Reperfusion Rats

摘要(Abstract):
目的探究参芎化瘀胶囊对脑缺血/再灌注(I/R)大鼠的保护作用及其机制。方法采用大脑中动脉线栓(MACO)法构建脑I/R模型大鼠,随机分为I/R模型组、参芎化瘀胶囊组(SHC)、尼莫地平组和MEK1/2激活剂TPA组,每组30只,另取30只作为假手术组。术后2 h,各组大鼠灌胃给予相应药物,TPA经脑室给药,连续5 d,每日1次;假手术组和模型组给予等体积含0.5%DMSO的0.9%氯化钠溶液灌胃处理。最后一次给药24 h后,采用Longa评分标准评价大鼠神经功能损伤程度,红四氮唑(TTC)法检测大鼠脑梗死体积,HE染色法观察脑组织形态学变化,AnnexinV-FITC/PI法检测神经细胞凋亡情况,蛋白印迹(WB)法检测脑组织中凋亡相关蛋白Bcl-2、Bax、caspase3表达及MAPK/ERK信号通路相关蛋白MEK1/2、p-MEK1/2、ERK1/2和pERK1/2表达。结果与模型组比较,SHC组大鼠神经功能评分明显降低(P <0.05),脑梗死体积明显减小(P <0.05),脑组织病变程度减轻,神经细胞凋亡指数显著降低(P <0.05),Bax、caspase3蛋白表达明显下调(P <0.05)、Bcl-2蛋白表达显著上调(P <0.05),TPA可抑制这一保护作用。与模型组比较,SHC组p-MEK1/2、pERK1/2蛋白表达均显著下调(P <0.05),可被TPA抑制。结论参芎化瘀胶囊通过抑制MAPK/ERK信号通路激活,抑制细胞凋亡,实现对脑缺血/再灌注神经损伤的保护作用。
