益气化瘀方对缺血-再灌注大鼠心肌细胞凋亡相关蛋白表达的影响Effect of Yiqi Huayu Decoction on the Expression of Apoptosis Related Proteins in Myocardial Cells of Rats with Ischemia Reperfusion Injury

摘要(Abstract):
目的观察益气化瘀方对缺血-再灌注(I/R)大鼠心肌细胞凋亡相关蛋白Bcl-2、Bax、Caspase-3、Fas及P53表达的影响。方法采用结扎左冠状动脉前降支,心肌缺血30 min,再灌注2、4 h制作缺血-再灌注模型。将大鼠随机分为7组,即假手术组、模型2 h组(缺血-再灌注2 h)、模型4 h组(缺血-再灌注4 h组)、通心络2 h组(缺血-再灌注2 h)、通心络4 h组(缺血-再灌注4 h)、益气化瘀方2 h组(缺血-再灌注2 h)、益气化瘀方4 h组(缺血-再灌注4 h)。造模前连续灌胃相应药物1周。免疫组化法测定Bcl-2、Bax、Caspase-3、Fas、P53蛋白表达。结果益气化瘀方可提高缺血-再灌注大鼠心肌Bcl-2蛋白表达量(P<0.05)、降低缺血-再灌注大鼠心肌Bax、Caspase-3、Fas、P53蛋白表达量(P<0.05)。结论益气化瘀方可减轻大鼠心肌缺血-再灌注细胞凋亡,其机理可能与上调Bcl-2蛋白表达和下调Bax、Caspase-3、fas、P53蛋白表达有关。
