炎调方对脓毒症大鼠肠黏膜屏障及TLR9信号通路影响的实验研究Effects of Yantiao Decoction on Intestinal Mucosal Barrier and TLR9 Signaling Pathway in Sepsis Rats

摘要(Abstract):
目的观察炎调方对脓毒症大鼠肠黏膜屏障及Toll样受体9(TLR9)信号通路的影响。方法将大鼠分为假手术组、模型组、地塞米松组及炎调方低、中、高剂量组,每组12只。进行盲肠结扎穿孔法建立脓毒症大鼠模型,术后24 h取材。苏木精-伊红染色(HE)观察各组大鼠小肠组织黏膜病理及通透性改变,进行Chiu评分,细菌移位率变化,检测大鼠血清肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)及小肠组织中二胺氧化酶(DAO)、肠型脂肪酸结合蛋白(i-FABP)水平、实时荧光定量(q RT-PCR)及蛋白免疫印迹法(Western blotting)检测TLR9、MyD88、核转录因子-κB(NF-κB)表达。结果与假手术组比较,模型组大鼠细菌移位率,小肠黏膜Chiu评分、TNF-α、IL-6水平、TLR9、MyD88、NF-κB表达显著升高,黏膜厚度、绒毛高度、DAO及i-FABP水平显著降低(P <0.05);与模型组比较,炎调方各剂量组和地塞米松组细菌移位率、Chiu评分、TNF-α、IL-6水平、TLR9、MyD88、NF-κB表达降低,黏膜厚度、绒毛高度及DAO、i-FABP水平升高,且炎调方各剂量组之间呈剂量依赖性(P <0.05);炎调方高剂量组和地塞米松组比较,差异无统计学意义(P> 0.05)。结论炎调方可能通过介导TLR9信号对脓毒症大鼠肠黏膜屏障起保护作用。
