调理中焦气机法对脓毒症大鼠肠道屏障损伤及炎症因子的影响Effects of Conditioning Zhongjiao Qi Mechanism on Intestinal Barrier Damage and Inflammatory Factors in Septic Rats

摘要(Abstract):
目的 观察调理中焦气机法对脓毒症大鼠肠道屏障损伤及炎症因子的影响。方法 SD大鼠采用盲肠结扎法建立脓毒症大鼠模型,随机分为模型组,中药高、中、低剂量组,每组各8只,另取SD大鼠8只仅做开腹处理作为假手术组,各组分别予对应药物灌胃。HE染色法观察大鼠组织病理学改变;酶联免疫吸附法(ELISA)检测血清内毒素、D-乳酸、二胺氧化酶及血清白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平;免疫组化法(IHC)检测大鼠肠组织中IL-1β、IL-6、TNF-α蛋白表达水平。结果 与假手术组比较,模型组黏膜层可见大面积上皮细胞脱落,固有层裸露,局部可见溃疡,伴有较多淋巴细胞浸润,局部可见静脉血管淤血扩张。与模型组比较,中药各干预组黏膜层损伤减轻,以高剂量组最轻,损伤评分差异有统计学意义(P <0.01)。与假手术组相比,模型组大鼠血清内毒素、D-乳酸、二胺氧化酶水平明显升高(P <0.01),血清IL-1β、IL-6、TNF-α水平均显著上升(P <0.01),肠组织IL-1β、IL-6、TNF-α蛋白表达水平升高(P <0.05)。与模型组相比,中药各剂量组大鼠血清内毒素、D-乳酸、二胺氧化酶水平有不同程度的降低(P <0.05或P <0.01),血清IL-1β、IL-6、TNF-α水平明显降低(P <0.01),肠组织IL-1β、IL-6、TNF-α蛋白表达水平不同程度降低(P <0.05)。结论 调理中焦气机法可减轻脓毒症大鼠肠道屏障损伤,其机制可能与其抑制炎症因子IL-1β、IL-6、TNF-α释放有关。
