电针对大脑中动脉阻塞大鼠学习记忆功能及海马CA1区突触素的影响Effects of EA at Baihui(DU20) and Shenting(DU24) on Learning-memory Function and the Expression of Synaptophysin in Hippocampal CA1 in a Cerebral Ischmeia-reperfusion Rat Model

摘要(Abstract):
目的观察电针对大脑中动脉阻塞大鼠学习记忆功能及其对海马CA1区突触素(SYN)的影响,探讨电针改善学习记忆的可能机制。方法造模手术组对SD大鼠的左侧大脑中动脉采用改良Longa线栓阻塞法栓塞90 min后再灌注实施手术,采用Zea Longa评分将造模成功的12只大鼠随机分为模型和电针组各6只。电针组取穴百会、神庭,治疗7 d。采用小动物核磁共振成像分析系统(MiniMR-60 MRI system 7.0 T)对大鼠进行T2加权成像(T2-weighted image,T2WI)扫描;学习记忆能力的检测选择Barnes巴恩斯迷宫;免疫荧光标记法观察缺血侧海马CA1区SYN的表达。结果与模型组相比,电针组在干预7 d后,Zea Longa评分更低(P=0.040<0.05)、其左侧脑梗死区域的面积明显减少(P <0.01);巴恩斯迷宫行为学测试发现,电针组逃避潜伏期明显缩短(P <0.001);进入错误洞口次数显著减少(P <0.001);免疫荧光结果显示,假手术组海马CA1区SYN表达最好,镜下可见大量荧光着色细胞且分布密集;模型组大鼠SYN的表达较假手术组表达明显减少;电针组SYN表达较模型组显著增加,镜下观察到散在荧光着色细胞,分布较密集。结论电针神庭、百会穴可以加强大脑中动脉阻塞大鼠海马CA1区SYN的表达,改善其突触可塑性,进而改善其学习记忆能力。
