基于Hippo-YAP信号通路探讨桃红四物汤对BMSCs增殖与成骨分化的影响Effect of Taohong Siwu Decoction on the Proliferation and Osteogenic Differentiation of BMSCs Based on the Hippo-YAP Signaling Pathway

摘要(Abstract):
目的 分离培养大鼠骨髓间充质干细胞(BMSCs),运用桃红四物汤含药血清和Hippo信号通路抑制剂Verteporfin干预,进而观察BMSCs增殖和成骨分化的能力。方法 采用全骨髓贴壁法体外培养BMSCs,选用10倍等效剂量的桃红四物汤灌服大鼠制备桃红四物汤含药血清,采取0.9%氯化钠注射液灌胃大鼠制取空白组对照血清,将BMSCs分为3组:空白血清组、桃红四物汤含药血清组(THSWT组)以及桃红四物汤含药血清+Verteporfin组(THSWT+V组),使用CCK-8分析法分析4个时相点0、1、3、7 d细胞增殖活力;干预7 d后,进行ALP测定,观察骨向分化程度;处理21 d后,进行茜素红染色,观察钙积累,Western blotting法检测各组细胞成骨关键因子及Hippo通路关键蛋白表达。结果 THSWT组BMSCs细胞增殖能力明显高于THSWT+V组及空白血清组(P <0.01);THSWT组BMSCs成骨分化能力及钙盐沉积程度高于THSWT+V组及空白血清组;THSWT组中YAP蛋白表达低于空白血清组、TAZ蛋白表达高于空白血清组(P <0.05);THSWT组ALP、OCN、RUNX2、Osterix等成骨关键因子蛋白表达明显高于THSWT+V组及空白血清组(P <0.05)。结论 桃红四物汤可促进BMSCs的增殖和成骨分化能力,从而促进骨折愈合,其作用机制为调节Hippo-YAP信号通路,激活其下游关键节点YAP蛋白及TAZ蛋白,诱导BMSCs成骨分化基因转录。
