丹参素对急性胰腺炎大鼠肾损伤的保护作用及对NF-κB信号通路的影响The Protective Effect of Danshensu on Renal Injury in Rats with SNP and Its Effect on NF-κB Signaling Pathway

摘要(Abstract):
目的探讨丹参素对急性坏死性胰腺炎(ANP)大鼠肾损伤的保护作用及对核因子-κB (NF-κB)信号通路的影响。方法制备ANP模型大鼠,随机分为ANP组、奥曲肽组、丹参素低[5 mg/(kg·d)]、中[10 mg/(kg·d)]、高[20 mg/(kg·d)]剂量组,假手术组及ANP组给予等量0.9%氯化钠注射液灌胃。造模后24 h,检测各组大鼠肾功能,苏木素-伊红(HE)染色观察胰腺组织、肾组织病理学变化,试剂盒检测血清中白细胞介素-6(IL-6)、白细胞介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)水平,蛋白印迹(WB)法检测肾组织中NF-κB、cleaved-caspase3表达情况。结果与假手术组比较,ANP组大鼠肾功能显著下降,血液中血清肌酐(Scr)、血尿素氮(BUN)含量显著升高,胰腺组织、肾组织现细胞水肿、脱落、坏死等病理变化,组织病理评分明显升高(P <0.05),血清IL-6、IL-8、TNF-α含量显著升高(P <0.05),肾组织细胞凋亡率明显增高(P <0.05),NF-κB核转移水平明显增加(P <0.05),cleaved-caspase3水平显著增高(P <0.05)。与ANP组比较,丹参素低、中、高剂量组大鼠肾功能均明显恢复,胰腺组织、肾组织病理变化均显著改善,组织病理评分显著降低(P <0.05),血清IL-6、IL-8、TNF-α含量显著下降(P <0.05),肾组织细胞凋亡率明显下降(P <0.05),NF-κB蛋白核转移水平明显减少(P <0.05),cleaved-caspase3水平显著降低(P <0.05),均呈剂量依赖效应。结论丹参素具有保护肾功能损伤的功能,其机制可能通过抑制NF-κB通路的激活,进而抑制细胞凋亡而实现。
