蒲参胶囊对大鼠脑缺血/再灌注后GAP-43和β-TubulinⅢ蛋白表达的影响

摘要(Abstract):
目的观察蒲参胶囊对大鼠脑缺血/再灌注后梗死区周边组织生长相关蛋白43(GAP-43)及β微管蛋白Ⅲ(β-TubulinⅢ)表达的影响。方法将健康SD大鼠随机分成蒲参胶囊低、中、高剂量组,假手术组和模型组。通过线栓法建立大鼠大脑中动脉阻塞(MCAO)脑缺血再灌注模型,术后高剂量组(250 mg/200 g)、中剂量组(125 mg/200 g)、低剂量组(62.5 mg/200 g)每日予灌胃给药1次,假手术、模型组给予0.9%氯化钠注射液灌胃。分别在术后第1、7、14日行神经功能缺损评分(NSS),用Western blotting检测梗死周边区GAP-43和β-TubulinⅢ的蛋白表达情况。结果通过NSS评分,蒲参胶囊对脑缺血/再灌注大鼠的神经功能损伤的恢复有一定的改善趋势,Western blotting检测显示,与模型组比较,蒲参胶囊中剂量组在术后7 d时,可提高GAP-43和β-TubulinⅢ蛋白水平(P <0.05或P <0.05);中剂量组在术后14 d,β-TubulinⅢ的蛋白水平也处于明显升高状态(P <0.05)。结论蒲参胶囊在一定程度上可提高GAP-43及β-TubulinⅢ的蛋白表达水平,提示蒲参胶囊具有促进脑缺血损伤后的神经再生修复的作用。
