“炎调方”调节晚期糖基化终末产物受体及其配体对脓毒症急性肺损伤大鼠保护作用Protective Effects of Yantiao Decoction on Sepsis-induced Acute Lung Injury in Rats by Regulating RAGE and Ligands

摘要(Abstract):
目的以CLP大鼠为模型,观察"炎调方"调节糖基化终末产物受体(RAGE)及其配体S100A12表达,探讨其对大鼠脓毒症急性肺损伤的保护作用。方法将50只雄性SD大鼠随机分为对照组、生理盐水组、炎调方组、假手术组。采用盲肠结扎穿孔法制备大鼠脓毒症急性肺损伤模型,并于造模后6、12 h予以相应药物灌胃。24 h后观察大鼠肺部病理改变;RT-PCR法检测RAGE及其配体S100A12 mRNA表达。ELISA法检测大鼠血浆sRAGE、S100A12及肿瘤坏死因子-α(TNF-α)水平,并作相关性分析。结果 SD大鼠CLP造模后大鼠肺部RAGE mRNA、S100A12 mRNA;外周血sRAGE、S100A12、TNF-α水平较对照组、炎调方组及假手术组显著升高(P <0.01);造模大鼠经"炎调方"灌胃后上述指标均显著下降(P <0.01)。大鼠肺部RAGE及其配体S100A12 mRNA表达与外周血sRAGE及S100A12水平具有显著正性相关性(相关系数RS>0.5)。结论"炎调方"可下调CLP诱导的脓毒症急性肺损伤大鼠肺部RAGE、S100A12 mRNA表达,从而降低大鼠外周血sRAGE、S100A12及TNF-α水平,可能是通过该作用机制抑制肺部炎症反应,从而发挥保护大鼠脓毒症急性肺损伤作用。
