溃结宁膏穴位敷贴对溃疡性结肠炎(脾肾阳虚证)大鼠Th17/Treg平衡的影响Effect of Point Application with Kuijiening Plaster on the Balance of Th17/Treg in Ulcerative Colitis Rats with Spleen-kidney Yang Deficiency

摘要(Abstract):
目的观察溃结宁膏穴位敷贴对溃疡性结肠炎(UC)(脾肾阳虚证)大鼠白介素-17(IL-17)及叉头状转录因子3(Foxp3)水平的影响。方法将40只SD大鼠随机分为正常对照组、溃疡性结肠炎(脾肾阳虚证)模型组(模型组)、溃结宁膏穴位敷贴治疗组(敷贴组)、柳氮磺吡啶治疗组(SASP组)。建立溃疡性结肠炎(脾肾阳虚证)大鼠模型,造模结束后治疗28 d,观察大鼠一般情况,进行疾病活动指数(DAI)评分、结肠黏膜损伤指数(CMDI)评分、组织学损伤指数(TDI)评分,酶联免疫吸附(ELISA)检测各组大鼠血清及结肠组织IL-17、Foxp3水平。结果与正常对照组大鼠比较,模型组大鼠体质量下降、饮水量减少、肛温降低,大鼠疾病活动指数评分、结肠黏膜损伤指数评分及结肠组织学损伤指数评分均显著升高,大鼠IL-17水平明显增高,Foxp3水平明显降低(P<0.01);与模型组大鼠相比较,敷贴组、SASP组大鼠体重增加、饮水量增加、肛温升高,大鼠疾病活动指数评分、结肠黏膜损伤指数评分及结肠组织学损伤指数评分明显降低,大鼠IL-17水平明显降低,Foxp3水平明显增高(P<0.01);敷贴组、SASP组之间差异无统计学意义(P>0.05)。结论溃结宁膏穴位敷贴可减轻UC肠道炎症反应,对肠道黏膜具有修复作用,其部分作用机制是通过上调抑炎因子Foxp3并下调促炎因子IL-17的水平,恢复Th17/Treg平衡来达到治疗UC的目的。
