长蛇灸激活PPARγ改善初发强直性脊柱炎大鼠的机制研究Mechanism of Long Snake Moxibustion Activation of PPARγ to Improve the Initial Onset of Ankylosing Spondylitis in Rats

摘要(Abstract):
目的 探讨长蛇灸通过激活过氧化物酶体增殖激活受体(PPARγ)来抑制炎性细胞因子表达从而缓解强直性脊柱炎(AS)的机制。方法 以3月龄大鼠为模型,用人蛋白多糖提取物溶于二甲基十八胺2 mg腹腔注射,每隔2周注射1次,共4次,构建强直性脊柱炎模型。在模型建立初期开始治疗干预,即用长蛇灸治疗2周,同时评估AS大鼠模型的严重程度。治疗结束后,采用ELISA法检测炎症细胞因子如白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-17(IL-17)和白细胞介素-23(IL-23)蛋白变化,qPCR检测IL-6、TNF-α、IL-1β、IL-17和IL-23的m RNA变化,Western blotting法检测PPARγ激活程度。结果 在AS初期即用长蛇灸治疗可显著下调AS大鼠外周病进展评分(P <0.05),且可显著抑制外周血TNF-α、IL-1、IL-17、IL-23和IL-6的蛋白和m RNA表达水平(P <0.05),同时激活组织中PPARγ的表达(P <0.05)。结论 在AS大鼠初期采用长蛇灸治疗可能通过激活PPARγ来抑制IL-6、IL-1β、TNF-α、IL-17和IL-23的表达,最终缓解强直性脊柱炎。
